HER2 is activated in two different ways
Overexpression:

Arising from gene amplification which results in
constitutive activation of HER2 (ligand independent)
o
Antagonized by trastuzumab
Co-receptor:
o
For other HER family members especially HER3
(ligand dependent)
o
Antagonized by pertuzumab
